Here are two way I know to do this:

First, using the dosfilelookup control template add the following in the
init embed priority 8800.  Where FileLookup1 is the object name of your file
lookup object.
  FileLookup1.Flags = BOR(FileLookup1.Flags,FILE:Directory)

Second, you can use the FileDialog command and set the directory flag.
